Abstract
Triazolopiperazines are medicinally relevant scaffolds with considerable potential for drug discovery. This work presents a modular, one-pot synthetic approach combined with acoustic droplet ejection and high-throughput experimentation. Sequential design of experiments was used to optimize reaction conditions, followed by a 1,536-reaction campaign to systematically explore chemical space. The resulting dataset provides a foundation for understanding structure–reactivity relationships and guiding future analogue synthesis.
